While the role of Computer-Aided Drug Design in small molecule pharmaceutical research appears to be well established, there are not that many accounts of its real contributions to specific therapeutic projects. In this presentation, we report a number of examples where computational methods were prospectively employed to successfully impact Roche research projects. We conclude by sharing the collective principles that guide us in productively employing computational approaches and highlight some of the technological developments we are undertaking to complement our capabilities.
